Abstract

The epigenetics is the study of changes in gene expression or celluar phenotype caused by mechanisms other than changes in the underlying DNA sequence. Epigenetic regulation in asthma and allergic diseases is an interesting area that has obtained a great deal of scientific concern in recent years. Allergic diseases such as asthma and allergic rhinitis in early life are one of the important indicators that the developing immune system is vulnerable to biologic and environmental pollutants. The epigenetic studies are likely to provide clear and deep understandings into how the environment can be modified to drive immune development and change the allergic disease phenotypes. This review includes 1) definition of epigenetics 2) regulation of epigenetics 3) characterization of environmental asthma and allergic diseases triggers that induce epigenetic changes.

CpG island (stretch of DNA > 200 bp in length with > 50% GC content, observed/expected CpG > 0.6)

또한 흡연은 p16 (cyclin-dependent kinase inhibitor 2A [melanoma, p16, inhibits CDK4; INK4a]), cytochrome P450, family 1, subfamily A, polypeptide 1 (CYP1A1) [46], Ras association(RalGDS/AF-6) domain family member 1 (RASSF1A)[47], fragile histidine triad gene (FHIT) [46], monoamine oxidase type B (MAOB) [48] 등의 후생유전변화에 관여한다.

Mechanism of epigenetic modulation [4]
Site (s) of action Mechanism (s) of action Effect (s)
DNA methylation CpG islands (stretches of DNA > 200 bp with > 50% GC content) and other CpG regions (< 200 bp) Addition of methyl group to the 5′ position of cytosines by DNMTs Turn off gene transcription
Histone modification Histone proteins (H2A, H2B, H3, and H4) that form the nucleosome core Chromatin remodeling by methylation, acetylation (via HAT, HDAC), phosphorylation, or ubiquitylation of histone tails Regulate accessibility of DNA to RNA polymerase II and transcription factors
Noncoding RNAs miRNAs (21-23 nucleotide long regulatory RNAs) Regulatory transcripts: binding of miRNA to the 3′ untranslated egions of mRNA Induce degradation of target mRNA
Table 3.
Asthma genes related to epigenetics [4]
Gene Mechanism Role in asthma
Il-4 Demethylation of an intronic sequence that binds GATA-3 Increases IL-4 secretion in TH lymphocytes
IL4 Increase in H3-K9 acetylation and H3-K4 trimethylation Increases lineage commitment of precursor TH cells to TH2 cells
Extensive demethylation of the 59 flanking region of the IL4 promoter Sustains high levels of IL-4 secretion from TH2 cells
Methylation of the 39 end of the IL4 locus Promotes differentiation of precursor TH1 cells into TH1 cells
IL5, IL13 Increased histone acetylation miR-21, miR-1 Increases TH2-associated cytokine expression
Overexpressed in IL13 transgenic mice
IL-12p35 miR-21 Downregulates gene expression in murine inflamed airway
IFN-γ Methylation of an activator protein 1-binding site in the proximal promoter resulting in reduced CREB (cAMP-responsive element binding protein 1 ) and Activating transcription factor 2 /c-Jun binding to this site Associated with the loss of gene expression and the establishment of a TH2 polarization phenotype
IFN- γ, CXCL10 Increased histone acetylation Inhibits TH1-associated recall responses and expression of these cytokines
TGFβ miR-146a Might mediate TGF-b plus cytomix-induced apoptosis
FOXP31 Class II HDAC inhibitors Increases Foxp31 expression and enhances the suppressive function of Foxp31 Treg cells on TH2 response
HLA-G miR-148a, miR-148b, and miR-152 Targets a single nucleotide polymorphism at the 39 untranslated region of the gene
POU domain class 2 associating factor 1 miR-126 Increases expression of the transcription factor
